AequoScreen Double Transfected Cell Line: Human recombinant 5-HT3A receptor in aequorin HEK-293 host cell.

We provide two vials of cryopreserved cells (approximately 2.5 x 106 cells/vial), detailed product information including cell line properties, culture conditions and the pharmacological properties of the recombinant channel in a functional assay.

All cell lines are tested for the absence of mycoplasma.

The AequoScreen technology is a generic GPCR technology which can be used with Gs, Gi and Gq coupled GPCRs and calcium coupled ion channels. Following receptor stimulation, increases in intracellular calcium enable measurement of the resulting flash luminescence signal.

AequoScreen aequorin-encoding Plasmids for performing aequorin luminescent assay starting from non-aequorin cell lines

Aequorin is a photo protein originating from the jellyfish Aequorea Victoria. The apo-enzyme (apoaequorin) is a 21 kD protein, which requires a hydrophobic prosthetic group, coelenterazine, to be converted to aequorin,the active form of the enzyme. This enzyme possesses 3 calcium binding sites which control its activity.

New Microplate Luminescence Counter for Aequorin based Ca2+-coupled GPCR Assays

The MicroBeta2 LumiJET introduces new features and possibilities for a variety of assays to be run with throughput and ease. The aequorin assays that are presented here were run with a 12-detector MicroBeta2 model with two injectors per detector. This is ideal for flash type luminescence assays as it combines high throughput with high sensitivity luminescence detection, therefore providing all tools for studying calcium related GPCR and ion channel assays.

Receptor-Mediated MAPK and Akt Signaling are Maintained in Gamma-Irradiated FroZen Cells

FroZen, -irradiated cells, are a well established product, that can be ordered as a consumable, and readily used to perform Aequorin functional assays (AequoZen) or cAMP assays (cAMPZen). While, for the G s–coupled receptors, an increase of cAMP is detected in such an assay, for G i-coupled receptors an inhibition of the forskolin-stimulated level of, cAMP is detected.
