The LANCE® Ultra Human BDNF Detection Kit is designed for detection and quantitation of human Brain Derived Neurotrophic Factor (BDNF) in buffered solution and cell culture media using a homogeneous TR-FRET (no-wash steps, no separation steps) assay.
LANCE® and LANCE® (Lanthanide chelate excite) Ultra are our TR-FRET (time-resolved fluorescence resonance energy transfer), homogeneous (no wash) technologies. One antibody of interest is labeled with a donor fluorophore (a LANCE Europium chelate) and the second molecule is labeled with an acceptor fluorophore (ULight™ dye). Upon excitation at 320 or 340 nm, energy can be transferred from the donor Europium chelate to the acceptor fluorophore if sufficiently close for FRET (~10 nm). This results in the emission of light at 665 nm.
Brain Derived Neurotrophic Factor (BDNF) is a neurotrophin that acts within the central and peripheral nervous systems. BDNF is expressed in a pro-form as pro-BDNF then later cleaved by plasmin, furin, or pro-convertase enzymes to become the mature form of BDNF. Released BDNF has been observed to bind to both TrkB and LNGFR (p75) receptors. Acting on these receptors, BDNF helps to support the differentiation of new neurons and synapses, promote neuron growth, and generally promote the survival of existing neurons. BDNF has been observed to potentially play a role in several major diseases such as Alzheimer’s, Parkinson’s and Huntington’s disease.
