Alpha-tubulin is a 451 amino acid protein that is one of the components of the microtubule cytoskeleton of eukaryotic cells. Alpha and beta tubulin form the skeleton itself, while gamma tubulin is involved in its organization. Alpha-tubulin is ubiquitous in every cell and tissue of the human body, and is highly preserved at the eukaryotic level (human 95% with mouse and 74% with yeast). As such, detection of alpha tubulin in cell extracts is a good tool for normalization of the amount of cell proteins per well in a biochemical assay. The alpha-tubulin AlphaLISA detection kit allows for the detection of alpha-tubulin in tissues homogenates and cell lysates.
AlphaLISA technology allows the detection of molecules of interest in a no-wash, highly sensitive, quantitative assay. In an AlphaLISA assay, a biotinylated anti-analyte antibody binds to the Streptavidin-coated Donor beads while another anti-analyte antibody is conjugated to AlphaLISA Acceptor beads. In the presence of the analyte, the beads come into close proximity. The excitation of the Donor beads causes the release of singlet oxygen molecules that triggers a cascade of energy transfer in the Acceptor beads, resulting in a sharp peak of light emission at 615 nm.
